近年来,登革热在全球的流行迅速增长,埃及伊蚊Aedes aegypti(L.)和白纹伊蚊Aedes albopictus(Skuse)是登革热传播的主要媒介。目前,化学防治仍然是防治蚊虫的主要手段,每年化学杀虫剂的大量使用直接或间接地导致其产生了抗药性。关于埃及伊蚊和白纹伊蚊的抗药性研究,主要集中于DDT、有机磷和拟除虫菊酯类杀虫剂导致的抗药性,并已深入到抗药性分子机理的研究。文章就埃及伊蚊和白纹伊蚊对杀虫剂诱导的抗药性现状及抗性机理的研究进展进行综述。 相似文献

The soil-water threshold range of chemical signals and reactive oxygen species (ROS) homeostasis could have a profound impact
on drought tolerance in wheat. A pot experiment was used to investigate the homeostasis between ROS and antioxidant defense
at five harvest dates, and its role in the correlation between soil-water threshold range of chemical signals and drought
tolerance in three wheat (Triticum aestivum) cultivars during progressive soil drying. The cultivars were bred at different periods, cv. BM1 (old), cv. Xiaoyan6 (recent),
and cv. Shan229 (modern). They were treated with progressive soil drying. Shoot biomass was affected by drought imposed by
two water treatments (90% and 55% field water capacity). The modern wheat cultivar had a lower ROS content and higher ROS-scavenging
antioxidant capacity with greater soil drying (68–25% soil water content) compared with the older cultivar. The modern cultivar
also had excellent adaptation to drought, with a longer survival of 22.7 days and less reduction in shoot biomass of 20.9%
due to early chemical signals and better balance between ROS production and antioxidants. The older cultivar had survival
of 15.3 days and 37.3% reduction of shoot biomass. A wider soil-water threshold range of chemical signals was positively correlated
with improved drought tolerance and better ROS homeostasis. These results suggest that ROS homeostasis acts as a regulator
in relationships between the soil-water threshold range of chemical signals and drought tolerance. 相似文献

To investigate the heterogeneous protein composition of highly polarized hepatocyte plasma membrane (PM), three PM-associated subfractions were obtained from freshly isolated rat hepatocytes using density gradient centrifugation. The origins of the three subfractions were determined by morphological analysis and western blotting. The proteins were subjected to either one-dimensional (1-D) SDS-PAGE or two-dimensional (2-D) benzyldimethyl-n-hexadecylammonium chloride (BAC)/SDS-PAGE before nano-Liquid Chromatography-Electrospray Ionization--tandem mass spectrometry analysis (LC-ESI-MS/MS). A total of 613 non-redundant proteins were identified, among which 371 (60.5%) proteins were classified as PM or membrane-associated proteins according to GO annotations and the literatures and 32.4% had transmembrane domains. PM proteins from microsomal portion possessed the highest percentage of transmembrane domain, about 46.5% of them containing at least one transmembrane domain. In addition to proteins known to be located at polarized liver PM regions, such as asialoglycoprotein receptor 2, desmoplakin and bile salt export pump, several proteins which had the potential to become novel subfraction-specific proteins were also identified, such as annexin a6, pannexin and radixin. Snake secreted phospholipasesA2 (sPLA2s) are widely used as pharmacological tools to investigate their role in diverse pathophysiological processes. Some members of snake venom sPLA2s have been found to block voltage-activated K(+) channels (K(v) channels). However, most studies involved in their effects on ion channels were indirectly performed on motor nerve terminals while few studies were directly done on native neurons. Here, a novel snake sPLA2 peptide neurotoxin, Natratoxin, composed of 119 amino acid residues and purified from Naja atra venom was reported. It was characterized using whole-cell patch-clamp in acutely dissociated rat dorsal root ganglion (DRG) neurons. It was found to effectively inhibit A-type K(+) currents and cause alterations of channel gating characters, such as the shifts of steady-state activation and inactivation curves to hyperpolarization direction and changes of V(1/2) and slope factor. Therefore, Natratoxin was suggested to be a gating modifier of K(v) channel. In addition, this inhibitory effect was found to be independent of its enzymatic activity. These results suggested that the toxin enacted its inhibitory effect by binding to K(v) channel. To further elucidate the structural basis for this electrophysiological phenomenon, we determined the crystal structure of Natratoxin at 2.2 A resolution by molecular replacement method and refined to an R-factor of 0.190. The observed overall fold has a different structural organization from other K(+) channel inhibitors in animal toxins. Compared with other K(v) channel inhibitors, a similar putative functional surface in its C-terminal was revealed to contribute to protein-protein interaction in such a blocking effect. Our results demonstrated that the spatial distribution of key amino acid residues matters most in the recognition of this toxin towards its channel target rather than its type of fold. 相似文献

初步确定了2个新的猪产仔数分子标记,雌激素受体基因ESR的第8外显子处的ESRB位点、催乳素受体基因的第7外显子的FSHRB位点。通过比较和分析多个产仔数的效应,初步确定了4个有利于产仔数提高的分子标记基因型,基因位点ESR、FSHRB的基因型BB的产仔数显著地高于AB、AA型;位点ESRB、PRLR的基因型AA的产仔数显著地高于AB、BB型;4个基因位点多态性与仔猪生长性能、母猪乳头数不存在显著的影响。 相似文献

2001—2002年度在广东东莞市选取不同的自然小生境,研究土壤肥力、土壤水分和生境郁闭度对薇甘菊(Mikania micrantha H.B.K)开花结实的影响,结果表明:土壤肥力较高,花数较多,花期较长,结实率较高,种子千粒重较大;但土壤肥力过高,虽然种子千粒重大,花期长,但花数少,结实率低。在开阔的生境中,薇甘菊花数多,花期长;在林荫处花期短,花数少,但种子千粒重反而有所升高;植株在郁闭度为10%-20%的生境中结实率最高,高于20%或低于10%结实率均有所降低,过强或过弱的光照均不利于结实。土壤湿度虽然对种子千粒重无明显影响,但对薇甘菊花数、花期和结实率的影响显著,土壤湿度大,花数多,结实率高,花期长。在自然生境中,三种因素的影响既相对独立又相互制约。 相似文献

在美国柽柳(Tamarix L.)被列为臭名昭著的十大我来杂草之一,而在中国,它则作为一种濒危物种生存状况堪忧。对多枝柽柳(Tamarix ramosissima L bd.)气体交换进行研究,探讨其蒸汽压差(VPD)、叶水势、气孔导度(g)和水分利用效率(WUE)之间的联系,结果表明,柽柳之所以能在沙漠生存又能在美国河岸地带疯狂扩展,主要原因可能是:①柽柳属地下水湿生植物,发达的根系能直达地下水。②柽柳适应性生境很广,作为一种阳性树种,柽柳非常耐旱,能够在很低的叶水势(φ)条件下(-4.59Mpa)进行光合作用,而较低的光补偿点和暗呼吸速率又反映出其耐阴湿的特点。③灵敏的气孔适应性调整:其g对VPD的灵敏性随着干湿季其体内的水分状况变化而变化,调整蒸腾速率,从而影响其WUE;gs与φ成显著正相关(P≤0.05);gs与WUE显著相关(P≤0.05),但只有当gs处于正在下降的时候,光合作用和蒸腾作用两个交换过程受轻微限制时,可得到水消耗和CO2吸收的最优协调,WUE达到最大值。这些适应性调整可能是柽柳能在美国河岸地带疯狂扩展的主要原因。实验结果表明,柽柳完全能够适应荒漠生长环境,因而导致柽柳在我国处于濒危状况的根本原因不是传统认为的干旱的自然环境,而主要是人为因素造成地下水位下降以及过度砍伐所致。 相似文献

The PD-L1 overexpression is an important event of immune escape and metastasis in triple-negative breast cancer (TNBC), but the molecular mechanism remains to be determined. Interferon gamma (IFNγ) represents a major driving force behind PD-L1 expression in tumor microenvironment, and histone deacetylase 2 (HDAC2) is required for IFN signaling. Here, we investigated the regulation of HDAC2 on the IFNγ-induced PD-L1 expression in TNBC cells. We found the HDAC2 and PD-L1 expression in TNBC was significantly higher than that in non-TNBC, and HDAC2 was positively correlated with PD-L1 expression. HDAC2 promoted PD-L1 induction by upregulating the phosphorylation of JAK1, JAK2, and STAT1, as well as the translocation of STAT1 to the nucleus and the recruitment of STAT1 to the PD-L1 promoter. Meanwhile, HDAC2 was recruited to the PD-L1 promoter by STAT1, and HDAC2 knockout compromised IFNγ-induced upregulation of H3K27, H3K9 acetylation, and the BRD4 recruitment in PD-L1 promoter. In addition, significant inhibition of proliferation, colony formation, migration, and cell cycle of TNBC cells were observed following knockout of HDAC2 in vitro. Furthermore, HDAC2 knockout reduced IFNγ-induced PD-L1 expression, lymphocyte infiltration, and retarded tumor growth and metastasis in the breast cancer mouse models. This study may provide evidence that HDAC2 promotes IFNγ-induced PD-L1 expression, suggesting a way for enhanced antitumor immunity when targeting the HDAC2 in TNBC.Subject terms: Breast cancer, Immune evasion 相似文献
